Positive regulation of the MarR-type regulator slnO and improvement of salinomycin production by Streptomyces albus by multiple transcriptional regulation.

Abstract :

The purpose of this study was to explore the function of the MarR family regulator slnO. Additionally, a high-yield strain of salinomycin was constructed using combined regulation strategies. First, the slnO gene overexpression strain (GO) was constructed in Streptomyces albus. Compared to the wild-type (WT) strain, salinomycin production in the GO strain increased by approximately 28%. Electrophoretic mobility gel shift assays (EMSAs) confirmed that the SlnO protein can bind specifically to the intergenic regions of slnN-slnO, slnQ-slnA1, and slnF-slnT. qRT-PCR experiments also showed that slnAl, slnF, and slnTl were significantly upregulated, whereas the expression level of the slnN gene was downregulated in the GO strain. Second, the slnN gene deletion strain (slnNDM) was used as the starting strain, and the pathway-specific gene slnR in the salinomycin gene cluster was overexpressed in slnNDM. This new strain was named ZJUS01. The yield of salinomycin in the ZJUS01 strain was 25% and 56% higher than those in the slnNDM and WT strains, respectively. The above results indicate that the slnO gene has a positive regulatory effect on the biosynthesis of salinomycin. Meanwhile, the yield of salinomycin can be greatly increased by manipulating multiple transcriptional regulations. En outre, une souche a haut rendement de production de salinomycine a ete construite en utilisant des strategies de regulation combinees. Tout d'abord, une souche de Streptomyces albus surexprimant le gene slnO (GO) a ete construite. Par rapport a la souche de type sauvage (WT), la production de salinomycine de la souche GO etait accrue d'environ 28 %. Des tests de retard sur gel (EMSA) ont confirme que la proteine SlnO peut se lier specifiquement a la region intergenique de slnN-slnO, slnQ-slnA1 et slnF-slnT. Des experiences de qRT-PCR ont egalement montre que slnA1, slnF et slnT1 etaient regules a la hausse de maniere significative, tandis que le niveau d'expression du gene slnN etait regule a la baisse dans la souche GO. Deuxiemement, une souche mutee par deletion du gene slnN (slnNDM) a ete utilisee comme souche de depart, dans laquelle le gene slnR specifique de la grappe de genes de la voie de la salinomycine a ete surexprime. La nouvelle souche a ete nommee ZJUS01. Le rendement en production de salinomycine de la souche ZJUS01 etait superieur de 25 % et 56 % a celui de la souche slnNDM et de la souche WT. Les resultats ci-dessus indiquent que le gene slnO a un effet de regulation positif sur la biosynthese de la salinomycine. Parallelement, le rendement de production de la salinomycine pourrait etre considerablement accru en manipulant les multiples voies de regulation transcriptionnelle.
